Anti-Integrin Alpha V + Beta 5抗體,整合素αVβ5抗體產(chǎn)品介紹：Integrins are heterodimeric proteins made up of alpha and beta subunits. At least 18 alpha and 8 beta subunits have been described in mammals. Integrin family members are membrane receptors involved in cell adhesion and recognition in a variety of processes including embryogenesis, hemostasis, tissue repair, immune response and metatastatic diffusion of tumour cells. ITAGV encodes integrin alpha chain V. Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. The I-domain containing integrin alpha V undergoes post-translational cleavage to yield disulfide-linked heavy and light chains, that combine with multiple integrin beta chains to form different integrins. Among the known associating beta chains (beta chains 1,3,5,6, and 8; ITGB1, ITGB3, ITGB5, ITGB6, and ITGB8), each can interact with extracellular matrix ligands; the alpha V beta 3 integrin, perhaps the most studied of these, is referred to as the Vitronectin receptor (VNR). In addition to adhesion, many integrins are known to facilitate signal transduction.

保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. 多抗的純化是一件比較簡單的事情，因為人工干預(yù)比較大，所以對實驗者的操作依賴性較強。很多新手覺得這是一件比較復(fù)雜困難的工作，但是如果理解了原理，就可以自行改進實驗了。多抗純化方法比較多，過去有用鹽析法純化的，有用辛酸-硫酸銨沉淀的方式純化的，也有用冷酒精沉淀的方式純化的，也有后來的離子交換層析和Protein A、G、A/G 純化的，但是在今天，市場上幾乎所有的抗體都是通過抗原親和純化得到的，與其它方法相比，抗原親和層析得到的抗體效率高、產(chǎn)量高、產(chǎn)品純、操作簡單。鑒于很多網(wǎng)友問起這方面的問題，我就詳細地講一講親和純化原理和操作。

說到親和層析，還得從上個世紀60年代說起。當(dāng)年美國NIH（National Institutes of Health）

有個學(xué)生在研究一種酶和抑制劑之間的相互作用的時候，突發(fā)奇想：既然此酶可以和抑制劑結(jié)合，那如果把抑制劑固定起來，不就可以用來純化酶了嗎？試了一下覺得果然可行于是發(fā)了論文，從那之后，就出現(xiàn)了各種各樣的親和層析，親和配基與配體可以是酶與底物、酶與抑制劑、激素與受體、抗原和抗體、糖蛋白與凝集素、*與親和素、*與二價金屬離子、卟啉環(huán)與亞鐵離子等。親和層析精髓：將一種分子固定，用來吸附另一種特異性分子，zui后將其洗脫下來。
