肝細(xì)胞生長(zhǎng)因子抗體保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

肝細(xì)胞生長(zhǎng)因子抗體,Anti-HGF抗體免疫應(yīng)答過(guò)程：

1.抗原進(jìn)入機(jī)體；

2.抗原刺激 B淋巴細(xì)胞，使之分化增殖成為抗體分泌漿細(xì)胞和記憶細(xì)胞（初次免疫應(yīng)答）；

3.抗體主要由漿細(xì)胞分泌 ；

4.記憶細(xì)胞再次受到特異性抗原刺激后會(huì)被激活而成為分泌抗體的漿細(xì)胞（再次免疫應(yīng)答）；

5.記憶B細(xì)胞是長(zhǎng)命細(xì)胞，可以存活數(shù)月甚至機(jī)體終生 。

Western Blotting 實(shí)驗(yàn)步驟概述

1、 組織取材：組織塊稱(chēng)重

2、 利用液氮、研缽粉碎組織塊

3、 加入RIPA緩沖液（每克組織3ml RIPA），PMSF（每克組織30μl，10 mg/ml PMSF），

利用Polytron進(jìn)一步勻漿（15,000轉(zhuǎn)/分*1分鐘）維持4℃

4、 加入PMSF（每克組織30μl，10 mg/ml PMSF），冰上孵育30分鐘

5、 移入離心管4℃，約20,000g（約15,000轉(zhuǎn)）15分鐘

6、 上清液為細(xì)胞裂解液可分裝-20℃保存

7、 進(jìn)行Bradford比色法測(cè)定蛋白質(zhì)濃度

8、 取相同質(zhì)量的細(xì)胞裂解液（體積*蛋白質(zhì)濃度），并加等體積的2×電泳加樣緩沖液

9、 沸水浴中3分鐘

10、上樣

11、電泳（濃縮膠20mA，分離膠35mA）

12、電轉(zhuǎn)膜儀轉(zhuǎn)膜（100mA 40分鐘）

13、膜用麗春紅染色，膠用考馬斯亮藍(lán)染色

14、Westernblot 試劑盒顯色

背景介紹：Hepatocyte growth factor regulates cell growth, cell motility, and morphogenesis by activating a tyrosine kinase signaling cascade after binding to the proto-oncogenic c-Met receptor. Hepatocyte growth factor is secreted by mesenchymal cells and acts as a multi-functional cytokine on cells of mainly epithelial origin. Its ability to stimulate mitogenesis, cell motility, and matrix invasion gives it a central role in angiogenesis, tumorogenesis, and tissue regeneration. It is secreted as a single inactive polypeptide and is cleaved by serine proteases into a 69-kDa alpha-chain and 34-kDa beta-chain. A disulfide bond between the alpha and beta chains produces the active, heterodimeric molecule. The protein belongs to the lasminogen subfamily of S1 peptidases but has no detectable protease activity. Alternative splicing of this gene produces multiple transcript variants encoding different isoforms.
