白介素1受體相關(guān)蛋白樣1前體蛋白抗體背景介紹：The protein encoded by this gene is a member of the interleukin 1 receptor family and is similar to the interleukin 1 accessory proteins. It is most closely related to interleukin 1 receptor accessory protein-like 2 (IL1RAPL2). This gene and IL1RAPL2 are located at a region on chromosome X that is associated with X-linked non-syndromic mental retardation. Deletions and mutations in this gene were found in patients with mental retardation. This gene is expressed at a high level in post-natal brain structures involved in the hippocampal memory system, which suggests a specialized role in the physiological processes underlying memory and learning abilities.

The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

2.可逆性:抗原與抗體結(jié)合形成抗原抗體復(fù)合物的過(guò)程是一種動(dòng)態(tài)平衡, 其反應(yīng)式為：Ag+AbAg·Ab；
3.適當(dāng)濃度和比例；
4.敏感性:在測(cè)定血清中某一物質(zhì)的含量時(shí)，化學(xué)比色法的敏感度為mg/ml水平，酶反應(yīng)測(cè)定法的敏感度約為5~10μg/ml，免疫測(cè)定中凝膠擴(kuò)散法的敏感度與酶反應(yīng)法相仿。
二、影響抗原抗體反應(yīng)的因素
1.電解質(zhì) ：抗原抗體特異結(jié)合后，膠體粒便可發(fā)生脫水作用，此時(shí)易受電解質(zhì)影響，如有適當(dāng)濃度的電解質(zhì)存在就會(huì)使它們失去部分負(fù)電荷而相互凝聚，于是出現(xiàn)明顯的凝集或沉淀現(xiàn)象。
2.酸堿度:合適的pH為6-8。
3.溫度:37℃通常是抗原體反應(yīng)zui適溫度。

1、染色過(guò)強(qiáng)？

原 因：抗體濃度過(guò)高或孵育時(shí)間過(guò)長(zhǎng)

解決方法：降低抗體滴度、抗體孵育時(shí)間：室溫1小時(shí)或4度過(guò)夜

原 因：孵育溫度過(guò)高超過(guò)37度

解決方法：一般在室溫20-28度

原 因：DAB顯色時(shí)間過(guò)長(zhǎng)或濃度過(guò)高
