人類免疫缺陷病毒2型抗體保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

抗原修復(fù)，必須注意以下問題：
1、修復(fù)液的選擇。
0.01M枸櫞酸（pH6.0）：應(yīng)用zui多的抗原修復(fù)液，適用于大多數(shù)的抗原，用該液修復(fù)后的抗原表達增強，抗原的定性和定位很理想，結(jié)果不錯;
0.05M EDTA（pH8.0）：應(yīng)用較多的抗原修復(fù)液，修復(fù)能力較強，對于保存時間較長的切片或目的蛋白表達較弱的組織有很好的修復(fù)效果，需要控制好修復(fù)時間，否則容易出現(xiàn)較重的背景;
0.01M TBS（pH7.4）：中性修復(fù)液，適用于大多數(shù)抗原，對于核定位蛋白有較好的修復(fù)效果;
*：應(yīng)用較多的抗原修復(fù)液，適用于大多數(shù)的抗原，使用溫度37℃，對于容易脫片的切片有很好的保護作用。

2.抗原熱修復(fù)時應(yīng)選擇的溫度。
據(jù)實驗認為溫度在92℃－98℃是合適的，尤其在95℃為，這是因為：（1）這種溫度未達沸騰，切片不容易脫離載玻片；（2）能夠解離和破壞與蛋白交聯(lián)的甲醛醛鍵等，處理時可以隨意選擇和確定抗原修復(fù)的作用時間。如果選擇高壓修復(fù)，因為溫度較高，時間不宜過長

3.抗原修復(fù)時有效溫度所需持續(xù)時間根據(jù)各單位的設(shè)備條件以及使用的儀器不同，抗原修復(fù)時在有效的溫度范圍內(nèi)所持續(xù)的時間也不一樣。
4.抗原修復(fù)液必須遵循自然降溫規(guī)律，否則效果不好或達不到抗原修復(fù)的目的。
5.盡量使用足量的抗原修復(fù)液，防止切片干涸。
6.切片必須附貼牢固，否則發(fā)生掉片。

人類免疫缺陷病毒2型抗體,Anti-HIV2 gp36抗體溶解方法:

方法1：我們的抗體（凍干粉）已經(jīng)包含了0.01M PBS、1% BSA和0.1% 防腐劑（疊氮鈉或*），您只需要加入滅菌的雙蒸水就行了?？贵w溶解后，置于-20℃保存，分裝后使用，避免反復(fù)凍融，可保證至少1年有效；

方法2：也可以只加入一半體積的雙蒸水，再用一半體積的甘油補足，置于-20℃保存，這樣抗體不會凍，有利于抗體的穩(wěn)定。

后續(xù)試驗時，再使用對應(yīng)的緩沖液稀釋成工作液，即用即配。

我們的抗體（凍干粉），在常溫放置下，至少一個月有效；若在-20℃下保存（*），至少三年有效。

Antibodies are prepared with 0.01M PBS, pH 7.4 with 1% BSA and 0.1% Sodium Azide, prior to lyophilization. Currently, we offer both lyophilized and reconstituted antibodies, both resulting in final concentration of 1mg/mL. We recommend two ways to reconstitute antibodies in lyophilized form:

1) Reconstituting by directly adding 100uL of sterile, distilled water.

2) Alternatively, if glycerol does not affect the experiment, add 50uL sterile distilled water, followed by adding 50uL of glycerol.

背景介紹：Human immunodeficiency virus type 2 (HIV2), orginally isolated from patients in West Africa, is the dominant form of HIV in West Africa capable of causing the acquired immunodeficiency syndrome (AIDS). HIV2 is closely related to simian immunodeficiency viruses (SIV). HIV1 and HIV2 share similarity in their genomes, transmission, clinical features, immunological effects, and in their action of binding to the same CD4 cellular receptor, but there are significant differences in the amino acid and nucleotide sequences of HIV1 and HIV2, especially within their envelope genes and proteins. Additionally, HIV2 may have a longer incubation period and may be less pathogenic than HIV1. HIV2 gp36 is a transmembrane protein located in the envelope of the virus specific to HIV2 that binds to the putative cellular receptor proteins P45 and P62.
