人免疫球蛋白G（IgG）ELISA檢測(cè)試劑盒標(biāo)本的保存 
一般說來，在 5 天內(nèi)測(cè)定的標(biāo)本可放置于 4℃，超過一周測(cè)定的需低溫凍存。 對(duì)收集后當(dāng)天進(jìn)行檢測(cè)的標(biāo)本，儲(chǔ)存在 4 ℃ 備用，如有特殊原因需要周期收集標(biāo)本， 將標(biāo)本及時(shí)分裝后放在 -20 ℃ 或 -70 ℃ 條件下保存。避免反復(fù)凍融。 標(biāo)本 2 -8 ℃ 可保存 48 小時(shí)， -20 ℃ 可保存 1 個(gè)月。 -70 度可保存 6 個(gè)月。部分標(biāo) 本也可加入適當(dāng)防腐劑，激素類標(biāo)本需添加抑肽酶。 

的基本原理
就是將抗原抗體固定到特定的載體上進(jìn)行反應(yīng)，并通過酶催化底物發(fā)生化學(xué)變化將抗原抗體反應(yīng)通過顏色的形式顯現(xiàn)出來。
（1）將抗原抗體反應(yīng)固相化：即試驗(yàn)中的包被環(huán)節(jié)，使抗原或抗體吸附于固相載體表面。其機(jī)制可能是聚苯乙烯表面間的疏水基團(tuán)可以無選擇性地吸附蛋白分子。此物理性吸附不破壞蛋白分子結(jié)構(gòu)，保持其生物學(xué)特性和免疫學(xué)活性；
（2）封閉：固相吸附蛋白是非特異性的，在包被目的免疫蛋白后，未吸附蛋白的固相表面仍然有吸附其它蛋白的能力，為了保障抗原抗體反應(yīng)的特異性，因此，包被剩余的固相表面應(yīng)該用抗原抗體反應(yīng)無關(guān)蛋白封閉。一般認(rèn)為牛血清白蛋白是zui為理想的封閉劑，也可用脫脂奶粉、明膠、牛血清替代。
（3）抗原抗體反應(yīng)：抗原或抗體在適當(dāng)?shù)慕橘|(zhì)下可進(jìn)行特異的抗原抗體反應(yīng)。 （4）酶反應(yīng)：酶可以通過共價(jià)鍵與抗原或抗體連接形成結(jié)合物，當(dāng)抗原抗體反應(yīng)的時(shí)候，被檢測(cè)靶目標(biāo)中也結(jié)合了酶分子。
（5）ELISA試劑盒底物反應(yīng)：結(jié)合在抗原抗體中的酶分子，可以促使底物發(fā)生顏色反應(yīng)，檢測(cè)人員可以裸眼觀察，可以通過顏色來判定是否有免疫反應(yīng)的存在，通過顏色的深淺判斷標(biāo)本中相應(yīng)抗原或抗體的含量，也可借用酶標(biāo)儀在適當(dāng)?shù)牟ㄩL(zhǎng)讀取吸光度值。
